This study aimed to conduct the survey of ostracofauna from Cantaure Formation (Early Miocene) and Paraguaná (Lower Pliocene) in Paraguaná Peninsula, Venezuela, and to take stock of existing genres that after the close connection between the Atlantic and Pacific oceans, contributing to scientific knowledge about Caribbean region. This is the first study specifically with ostracodes these formations. The material study consisted of a total of five samples, three from Cantaure and two from Paraguaná. Were found a total of 32 genera of ostracodes in Cantaure Formation and 13 in Paraguaná Formation. The difference in diversity between Cantaure and Paraguaná formations is probably linked with the final uplift of Panama Isthmus, which occurred in the Pliocene and it is associated with a drop in salinity and decrease the amount of nutrients available. The reduction of the fauna occurred in the interval between Miocene and Pliocene agree what was said by other authors for the Caribbean region. A fauna with 2.6% endemism in the lower Miocene has decreased to 1.2% in the Pliocene. Among the marine fauna associated were found carophytas and the ostracod genre Ilyocypris in Paraguaná Fm, which are indicators of freshwater to mixohalinos environments, suggesting that this peninsula was some influenced by a river. O soerguimento do Istmo do Panamá entre as Américas foi um dos eventos de maior importância ocorridos durante o Neógeno, uma vez que propiciou a união dos dois continentes após 50 milhões de anos de isolamento resultando na separação dos Oceanos Atlântico e Pacífico, e conseqüentemente em grandes mudanças, tanto físicas quanto químicas. O presente trabalho teve o objetivo de realizar o levantamento da ostracofauna das formações Paranaguá (Plioceno Inferior) e Cantaure (Mioceno Inferior) na Península de Paraguaná na Venezuela, bem como fazer o levantamento dos gêneros que se mantiveram após o fechamento da conexão existentes entre os oceanos Atlântico e Pacífico contribuindo para o conhecimento científico sobre essa região do Caribe, sendo esse o primeiro estudo realizado especificamente com ostracodes para essas formações. O estudo consistiu de um total de cinco amostras, sendo três da Formação Cantaure e duas da Formação Paraguaná, onde foram encontrados um total de 13 gêneros de ostracodes na Formação Paraguaná e 32 na Formação Cantaure, mostrando a diferença na proporção dos gêneros entre as duas formações, o que indica provavelmente a redução na diversidade da Formação Cantaure para a encontrada na Formação Paraguaná, a qual está ligada ao soerguimento final do Istimo do Panamá, ocorrida no Plioceno associada à queda da salinidade e diminuição da quantidade de nutrientes disponíveis. A redução da fauna ocorrida no intervalo entre Mioceno e Plioceno concorda o que foi dito por outros autores para a região do Caribe que de uma fauna com 2,6% de endemismo durante o Mioceno inferior sofreu uma diminuição para 1,2% no Plioceno.
