Nerium oleander Linn is a Apocynaceae shrub – it is among the most toxic ornamental plants, especially for children and animals – potentially able to lead to death if ingested. There are few studies relating N.oleander to neural and behavioral alterations in the current literature. The present study aim to describe novel effects of the Ethanolic Extract of N. oleander (EENO). Oleandrin was separated and confirmed by liquid-liquid extraction and analyzed by high performance liquid chromatography coupled to a diode array detector (HPLC-DAD), on a reversed phase C18 column and employing water-acetonitrile (63:37) (v/v) as a mobile phase. The EENO was injected by a dose: 150mg/kg i.p. to a total of 63 wistar rats divided into groups of 9 animals. The behavioral test demonstrated two phases of CNS activity, I-phase of myorelaxation and depression and the phase II with excitability, the two phases were verified by Electroencephalogram (EEG) and Electromyogram (EMG) of the animals. The outbreak of convulsions caused by the extract indicates the possibility of inclusion in the therapeutic protocol of anticonvulsive drugs such as diazepam, especially in people who are more susceptible as in epileptic patients.
