Best Aquatic Park located at the National Research Institute of the Amazon (LMA / Inpa). For this, analyzes were performed on fecal samples and rectal swab from 34 animals of different age groups, being nine adults, 12 puppies and 13 juveniles; in total 14 females and 20 males. The methodological process was carried out to fully characterize the strains, by isolating colonies characteristic of Salmonella spp .; identification of morphotintorial characteristics by Gram stain; identification of isolates according to the typical biochemical behavior of Salmonella spp. Triple Iron Sugar Agar (TSI), Iron Lysine Agar (LIA), Hydrogen Sulfide, Indole and Motility Agar (SIM) and Simmons Citrate Agar (CIT); confirmation of the isolates using the Polymerase Chain Reaction (PCR) technique; characterization of the sensitivity profile against the antimicrobial agents tetracyclines (30μg), ampicillin (10μg), gentamicin (10μg), cefoxitin (30μg), amoxicillin (30μg), ciprofloxacin (5μg), penicillin (10μg) and azithromycin (15μg); previous serotyping, carried out by slide agglutination tests, with specific antisera for Salmonella spp. and genetic isolation to confirm serotypes. The results of the comparison between the two collection methods were analyzed by Fisher's exact test using the GraphPad InStat program version 3.10. Of the eleven strains isolated through microbiological cultivation and biochemical characterization, nine showed positive results in PCR, confirming the positivity of three animals. The isolates came from samples collected from two young females and one juvenile female. Corresponding to a frequency of 16.6% in puppies and 7.7% in juveniles. The largest number of isolates was obtained from stool samples, however, due to the low number of positive samples, there was no significant difference between the two collection methods according to Fisher's exact test. According to the antibiogram, the isolated strains showed higher rates of efficacy for the use of tetracycline, gentamicin and ciprofloxacin. The strains were identified as Salmonella enterica subsp. salamae serotype II 42: r: - and Salmonella enterica subsp. enterica serotype Javiana. This being the first report of the occurrence of these serotypes in Amazonian manatees and the first report of Salmonella spp. in live sirenians. Objetivou-se investigar a frequência de Salmonella spp. em peixes-bois da Amazônia (Trichechus inunguis) mantidos em cativeiro no Parque Aquático Robin C. Best localizado no Instituto Nacional de Pesquisas da Amazônia (LMA/Inpa). Para isso, foram realizadas análises em amostras de fezes e swab retal de 34 animais de diferentes classes etárias, sendo nove adultos, 12 filhotes e 13 juvenis; no total de 14 fêmeas e 20 machos. O processo metodológico foi realizado para caracterização ampla das cepas, mediante o isolamento de colônias características de Salmonella spp.; identificação das características morfotintoriais pela coloração de Gram; identificação dos isolados conforme o comportamento bioquímico típico de Salmonella spp. em Ágar Ferro Triplo Açúcar (TSI), Ágar Lisina Ferro (LIA), Ágar Sulfeto de hidrogênio, Indol e Motilidade (SIM) e Ágar Citrato de Simmons (CIT); confirmação dos isolados com o uso da técnica de Reação em Cadeia da Polimerase (PCR); caracterização do perfil de sensibilidade frente aos agentes antimicrobianos tetraciclinas (30μg), ampicilina (10μg), gentamicina (10μg), cefoxitina (30μg), amoxicilina (30μg), ciprofloxacina (5μg), penicilina (10μg) e azitromicina (15μg); sorotipificação prévia, realizada por testes de aglutinação em lâmina, com anti-soros específicos para Salmonella spp. e isolamento genético para confirmação dos sorotipos. Os resultados da comparação entre os dois métodos de coletas foram analisados pelo teste exato de Fisher empregando-se o programa GraphPad InStat versão 3.10. Das onze cepas isoladas por meio do cultivo microbiológico e caracterização bioquímica, nove apresentaram resultados positivos na PCR, confirmando a positividade de três animais. Os isolados eram provenientes de amostras coletadas em duas fêmeas filhotes e uma fêmea juvenil. Correspondendo a frequência de 16,6% em filhotes e 7,7% em juvenis. O maior número de isolados foi obtido a partir de amostras de fezes, no entanto, devido ao baixo número de amostras positivas, não houve diferença significativa entre os dois métodos de coleta conforme o teste exato de Fisher. Conforme o antibiograma as cepas isoladas apresentaram maiores índices de eficácia ao uso de tetraciclina, gentamicina e ciprofloxacina. As cepas foram identificadas como Salmonella enterica subsp. salamae sorotipo II 42:r:- e Salmonella enterica subsp. enterica sorotipo Javiana.
