Hypostomus melanephelis, new species, is described from the rio Tapajós basin, Pará State, Brazil. It is distinguished from all congeners by having conspicuous, roundish and tiny set of dark spots, abdominal region mostly naked, a depressed head, keels along dorsal, mid-dorsal and median lateral series of plates, and a caudal fin usually without spots.
